Ten Commandments of DNA and Polyphosphate Enzymology PDF Print E-mail
Lessons learned from forty years of work on DNA replication and the last ten years on inorganic polyphosphate will be updated in the context of ten THOU SHALTs:
I. rely on enzymology to clarify biologic questions;
II. Trust the universality of biochemistry and the power of microbiology;
III. do not believe something because you can explain it;
IV. do not waste clean thinking on dirty enzymes;
V. do not waste clean enzymes on dirty substrates;
VI. depend on viruses to open windows;
VII. correct for extract dilution with molecular crowding;
VIII. respect the personality of DNA;
IX. use reverse genetics and genomics;
X. employ enzymes as unique reagents.

These several topics will be presented in a historical and personalized context.
